DNA sequence analysis of crown gall tumor T-DNA encoding the 0.7 kb transcript.
نویسنده
چکیده
Crown gall tumor formation involves integration into the plant genome of DNA sequences (the T-region) of tumor-inducing (Ti) plasmids present in Agrobacterium tumefaciens. The T-DNA of the tumor expresses several gene products. Little is known about the function or regulation of expression of the 0.7kb transcript, which represents a relatively abundant T-DNA transcript in octopine-type tumors. In this report, a detailed structural analysis of the gene encoding the 0.7 kb transcript has been obtained by DNA sequence analysis of T-DNA isolated from A6S/2 tumor line. An indication of the structural characteristics of the protein product is obtained from the predicted amino acid sequence. The sequences flanking the open reading frame show characteristics with other eucaryotic genes. The corresponding DNA sequence of the inducing Ti plasmid (pTiA6) is identical with that of the DNA sequence from the tumor. Comparison of this gene sequence with the corresponding region of another Ti plasmid (pTiAch5) shows several differences in the 5' flanking sequence, but the nucleotide sequence of the coding region and 3' flanking region are identical.

Unusual plasmid DNA organization in an octopine crown gall tumor.
A cloned tobacco crown gall tumor, 1595501, incited by A. tumefaciens strain 15955 was studied. Molecular analysis of the organization of the T-DNA by means of Southern transfer and hybridization techniques indicated that the 1595501 tumor has about 10 copies of TL DNA, five of which are complete TL DNA, whereas most octopine tumors have only one to two copies of complete TL DNA. Hybridization ...
متن کاملAgrobacterium tumefaciens DNA and PS8 bacteriophage DNA not detected in crown gall tumors.
Renaturation kinetics of labeled Agrobacterium tumefaciens DNA are not influenced by addition of 10(4)-fold excess of crown gall tumor DNA. Reconstruction experiments demonstrated that 0.01% added bacterial DNA produces a detectable increase in rate of renaturation of labeled DNA. Crown gall tumor DNA therefore cannot contain as much as 0.01% A. tumefaciens DNA (one entire bacterial genome per ...

To carry out this study, total DNA was extracted from eggs and from second stage juveniles of several populations of Meloidogyne javanica and Meloidogyne incognita, using phenol / chloroform method. Following extraction, DNA was electrophoresed on 1% agarose gel to determine its quality and quantity. A specific primer pair (C2F3 / 1108 23 and 20 nucleotides, respectively) was used to discrimina...

متن کاملDNA Methylation Mediated Control of Gene Expression Is Critical for Development of Crown Gall Tumors
Crown gall tumors develop after integration of the T-DNA of virulent Agrobacterium tumefaciens strains into the plant genome. Expression of the T-DNA-encoded oncogenes triggers proliferation and differentiation of transformed plant cells. Crown gall development is known to be accompanied by global changes in transcription, metabolite levels, and physiological processes. High levels of abscisic ...
